Επίδραση της ποιότητας και της ποσότητας του πρωτογάλατος στη διάρροια των μόσχων λόγω μόλυνσης με Cryptosporidium spp. (Master thesis)

Neonatal calf diarrhea is a syndrome of young calves aged less than 1 month old, provoked by the combination of infectious and non infectious causes. Cryptosporidium spp. as well as rotaviruses are the most commonly found causes in cases of diarrheic fecal samples. Colostrum management is the most important preventive measure, leading to adequate passive transfer to the newborn calves and thus, to the reduction of the neonatal diarrhea, including Cryptosporidium spp. The aim of this study was to assess the effect of colostrum quality and quantity on Cryptosporidium spp. diarrhea, during different seasons of the year, in an intensive dairy cattle farm, in Greece. Fecal samples were, randomly, collected from 100 calves, including all seasons of the year 2016. Screening for Cryptosporidium spp. oocysts was performed by staining fecal smears with Ziehl-Nielsen stain. At the time of collection, the consistency of each fecal sample was recorded according to the following manner: (1) normal feces, (2) soft feces and (3) watery feces. Colostrum quality was evaluated for each cow (with a calf included in the study) directly after the parturition, with the help of a colostrum meter. The total volume of the two colostrum meals, fed during the first 24 hours, was recorded using a graduated measuring cylinder. Both parametric and non-parametric statistical methods were applied for the statistical evaluation of the data. All analyses were conducted using the statistical software program SPSS. In total, 71% calves were positive for Cryptosporidium spp. oocysts. The statistical analysis revealed influence of colostrum quality on Cryptosporidium spp. oocysts shedding through feces. Linear regression showed that the colostrum quantity, during the first two meals post partum, was negatively associated with the number of Cryptosporidium spp. oocysts in feces. During multivariable analysis, the variables representing the quality of colostrum and the season of the calf’s birth were identified as confounders. The results of this study showed that the percentage of Cryptosporidium spp. infection was high among young dairy calves of the herd, leading to high risk of diarrhea. Colostrum management (colostrum quality and quantity) and season of the calf’s birth influenced the number of Cryprosporidium spp. oocysts. Our study proved that risk factors such as the colostrum quality and quantity, as well as the season, can affect the Cryptosporidium spp. shedding and the consistency of the feces. The above findings demonstrate novel risk factors that should be included in the strategic approaches to control cryptosporidiosis.

Abstract: Η διάρροια των μόσχων είναι ένα σύνδρομο των νεαρών (<1 μήνα) μόσχων που οφείλεται στο συνδυασμό λοιμωδών και μη λοιμωδών αιτιών. Το πρωτόζωο παράσιτο Cryptosporidium spp. αποτελεί ένα από τα πιο συχνά αίτια διαρροϊκών κοπράνων σε μόσχους, αυτής της ηλικίας. Η ορθή διαχείριση του πρωτογάλακτος συμβάλει στην επαρκή παθητική μεταφορά αντισωμάτων στους νεογέννητους μόσχους και κατά συνέπεια, στη μείωση των περιστατικών διάρροιας από Cryptosporidium spp. Σκοπός της παρούσας μελέτης ήταν να αξιολογηθεί η επίδραση της ποιότητας και της ποσότητας του χορηγούμενου πρωτογάλακτος έναντι της προκαλούμενης από Cryptosporidium spp. διάρροιας καθ’ όλη τη διάρκεια του έτους (διαφορετικές εποχές), σε μια ελληνική εκτροφή γαλακτοπαραγωγών αγελάδων, στην οποία εφαρμόζεται το εντατικό σύστημα εκτροφής. Δείγματα κοπράνων συλλέχθηκαν τυχαία από 100 μόσχους μίας εκτροφής, καθ’ όλη τη διάρκεια του έτους 2016. Ο έλεγχος της πιθανής ύπαρξης ωοκύστεων του πρωτόζωου παρασίτου Cryptosporidium spp. πραγματοποιήθηκε με την χρώση Ziehl-Nielsen σε επιχρίσματα κοπράνων. Κατά τη συλλογή τους, η σύσταση του κάθε δείγματος κοπράνων καταγράφηκε ως εξής: (1) κανονικά κόπρανα, (2) μαλακά κόπρανα και (3) υδαρή κόπρανα. Η ποιότητα του πρωτογάλακτος αξιολογήθηκε για κάθε αγελάδα, αμέσως μετά τον τοκετό, με τη βοήθεια ενός μετρητή πρωτογάλακτος (colostrum meter). Ο συνολικός όγκος των 2 πρώτων γευμάτων πρωτογάλακτος, καταγράφηκε με τη βοήθεια ενός βαθμονομημένου ογκομετρικού κυλίνδρου. Όλες οι στατιστικές αναλύσεις πραγματοποιήθηκαν με τη χρήση του λογισμικού SPSS (version 23.0). % Τα εξετασθέντα δείγματα κοπράνων σε ποσοστό 71% βρέθηκαν θετικά στο πρωτόζωο παράσιτο Cryptosporidium spp. Η στατιστική ανάλυση έδειξε επίδραση της ποιότητας του πρωτογάλακτος στον αριθμό των εν λόγω παρασίτων στα κόπρανα. Η γραμμική παλινδρόμηση έδειξε αρνητική συσχέτιση της ποσότητας του πρωτογάλακτος, κατά τη διάρκεια της πρώτης ημέρας της ζωής των μόσχων, με τον αριθμό των Cryptosporidium spp. στα κόπρανα. Τόσο η ποιότητα του πρωτογάλατος όσο και η εποχή γέννησης των μόσχων φάνηκε να αλληλεπιδρούν στον αριθμό των αποβαλλόμενων με τα κόπρανα ωοκύστεων του παρασίτου. Τα αποτελέσματα της παρούσας έρευνας έδειξαν ότι η μόλυνση με το πρωτόζωο παράσιτο Cryptosporidium spp., στη συγκεκριμένη εκτροφή, βρέθηκε σε υψηλά ποσοστά και η παρουσία του συνδέθηκε με μεγάλη πιθανότητα εμφάνισης διαρροϊκών κοπράνων. Η ορθή διαχείριση του πρωτογάλατος (ποιότητα-ποσότητα) και η εποχή του έτους επηρέασαν τον αριθμό των Cryprosporidium spp. καθώς και τη σύσταση των κοπράνων. Η μελέτη μας απέδειξε ότι παράγοντες κινδύνου, όπως η ποιότητα και η ποσότητα του πρωτογάλατος, καθώς και η εποχή, μπορούν να επηρεάσουν τον αριθμό των Cryprosporidium spp. καθώς και τη σύσταση των κοπράνων. Τα παραπάνω ευρήματα παρουσίασαν νέους παράγοντες κινδύνου, οι οποίοι θα πρέπει να συμπεριληφθούν στις στρατηγικές για τον έλεγχο και την πρόληψη της κρυπτοσποριδίωσης.
